Participants can perform computational processing of CryoEM images to produce a 3D density map of a specimen of interest from vitreous ice. Participants will understand the principles of parameter selection and will be able to apply their skills to unknown samples. CryoEM 3D Reconstruction is Module 5 in the 6-module series required to earn the advanced CryoEM Practitioner badge. The CryoEM Practitioner badge consists of six modules, each representing a fundamental skill essential to the practicing cryo-electron microscopist. Trainees may complete single modules to learn particular skills or earn the advanced CryoEM Practitioner badge by completing all modules. The modules are: 1) CryoEM Sample Preparation, 2) CryoEM Vitrification, 3) CryoEM Electron Microscopy, 4) Data Acquisition, 5) CryoEM 3D Reconstruction, and 6) CryoEM Visualization & Modeling.

Earning Criteria
Required
To earn each badge, a participant must receive a mark of “Strong” on 70% of rubric items and a mark no lower than “Adequate” on EVERY rubric item. To earn the advanced CryoEM Practitioner badge, all module badges must be completed.
